Accutase® Cell Detachment Solution contains proteolytic and collagenolytic enzymes for cell detachment. A sterile filtered ready-to-use solution specifically developed for the gentle and routine detachment of cells from standard tissue culture plasticware and adhesion coated plasticware.

Accutase performs exceptionally well in detaching primary and stem cells and maintaining high cell viability compared to animal origin enzymes such as trypsin. Some downstream applications following Accutase treatment include analysis of cell surface markers, virus growth assay, cell proliferation, tumor cell migration assays, routine cell passage, production scale-up (bioreactor), and flow cytometry. Cell lines tested for Accutase application include human embryonic stem cells, human mesenchymal stem cells, human neural stem cells and primary cells including macrophages, fibroblasts, keratinocytes, vascular endothelial cells, hepatocytes, and established cell lines such as adherent CHO and BHK cells and 293 cells. FAQs

Do I need to wash my cells before using Accutase?

No.  Accutase can be added once the media is removed.  After adding enough Accutase to cover your cells, simply wait for them to “ball up”. 

Do I need to stop the dissociation action of Accutase with serum?

Usually not.  Accutase is gentle enough that simple dilution of the reagent with DPBS or media is needed to stop the dissociation activity.  In unusual cases where inactivation is required, the standard trypsin inhibitors will work, such as Soybean Trypsin Inhibitor

How long should I leave Accutase on my cells?

Detachment times for Accutase are similar to trypsin, and the time should be determined empirically.  Observe the cultures under a microscope and watch for dissociation to occur.  In general, cells can be left in Accutase without damage for a much longer time than trypsin.

What is Accutase made from?

Accutase contains no mammalian or bacterial components.  It is a natural enzyme mixture with proteolytic and collagenolytic enzyme activity.  This means it mimics the action of trypsin and collagenase at the same time. However, because it is more efficient than mammalian trypsin or collagenase, it is formulated at a much lower concentration, making it less toxic and gentler on cells with the same effectiveness.

Is a bottle of Accutase that is partially thawed still OK to use?

Yes.  As long as the bottle still contains a remaining “ice cube” or is cool to the touch, it can be placed in the refrigerator for later use.  A partially thawed bottle can be refrozen for long term storage within 2 months of receipt.
